Abstract:

Non-small cell lung cancer (NSCLC) accounts for more than 80% of all lung cancer cases and ranks as the leading cause of cancer-related death worldwide. In the tumor microenvironment, dysregulated expression of inflammatory cytokines serves as a core driver of immunosuppression and therapeutic resistance, and such dysregulated cytokine networks further facilitate tumor initiation and progression. Based on the latest research advances on inflammatory cytokines and tumor progression, this review systematically summarizes the biological functions and regulatory mechanisms of nine key cytokines involved in NSCLC. Classified by their functional characteristics and biological roles, this review further elaborates the action mechanisms of interleukin-1β, interleukin-6, interleukin-8, interleukin-10, interleukin-12p70, interleukin-17, tumor necrosis factor-α, interferon-α and interferon-γ. In-depth analysis of the context-dependent regulatory patterns of these cytokines not only facilitates the identification of candidate biomarkers for predicting response and adverse events to immune checkpoint inhibitors, but also lays a theoretical foundation for the development of novel combination strategies targeting specific signaling axes.
